Phenol red is a pH indicator frequently used in cell biology laboratories.

What does phenol red do?

Phenol red is a water-soluble dye used as a pH indicator, changing from yellow to red over pH 6.6 to 8.0, and then turning a bright pink color above pH 8.1. As such, phenol red can be used as a pH indicator dye in various medical and cell biology tests.

What does the phenol red test test for?

What is this? Phenol Red Broth is a general-purpose differential test medium typically used to differentiate gram negative enteric bacteria. It contains peptone, phenol red (a pH indicator), a Durham tube, and one carbohydrate (glucose, lactose, or sucrose).

What Colour is phenol red?

Phenol red, also known as phenolsulfonphthalein, is a pH indicator dye that exhibits a gradual transition from yellow to red over a pH range of 6.2 to 8.2 (Figure 2). Above 8.2 the dye turns a bright fuchsia color.

Is phenol red acidic?

Phenol red exists as a red crystal that is stable in air. Its solubility is 0.77 grams per liter (g/l) in water and 2.9 g/l in ethanol. It is a weak acid with pK_a = 8.00 at 20 °C (68 °F). A solution of phenol red is used as a pH indicator, often in cell culture.

How do you make phenol red?

To prepare phenol red, use a 0.02% aqueous solution if phenol red is available or an alcoholic solution if phenol red, sodium salt is available. For the aqueous solution, add 0.02 g of phenol red to 75 ml of DI water. Dilute to a final volume 100 ml with DI water.

What does it mean when Phenol Red is orange?

Phenol red is yellow at a pH < 6.8 and red at a pH of > 7.4, therefore if a bacterium ferments a sugar to acid a yellow color will develop. ... Sometimes an orange color will develop in the tube, this represents a pH > 6.8 and should not be considered positive.

Why is Phenol Red used in culture media?

Phenol red is used as a pH indicator. The sodium salt of Phenol Red is widely used in culture media to identify changes of neutral to acidic pH values. A Phenol Red solution has: A yellow color at a pH of 6.4 or below.

What does methyl red test for?

The methyl red (MR) test detects the production of sufficient acid during the fermentation of glucose and the maintenance of conditions such that the pH of an old culture is sustained below a value of about 4.5, as shown by a change in the colour of the methyl red indicator which is added at the end of the period of ...

What does phenol red smell like?

Phenol has a distinct odor that is sickeningly sweet and tarry. You can taste and smell phenol at levels lower than those that are associated with harmful effects. Phenol evaporates more slowly than water, and a moderate amount can form a solution with water.

How do you take phenol red tablets?

What does phenol red contain?

We use three different kinds of phenol red broths. One contains glucose; one contains lactose, and the last contains sucrose. The objective of the exercise is to determine which organisms can utilize each sugar. Phenol red is a pH indicator which turns yellow below a pH of 6.8 and fuchsia above a pH of 7.4.

Is phenol red light sensitive?

Note that phenol red is a chromophore that will absorb light and thus decrease the effective illumination of a sample. More importantly, phenol red can contribute to background and can even mark lysosomes in cells illuminated with near UV light.

What happens when you mix baking soda calcium chloride and phenol red?

Description: This video shows a chemical reaction of sodium bicarbonate (baking soda), calcium chloride (road salt), and an indicator phenol red. The chemicals react to form calcium carbonate, sodium chloride, and carbon dioxide gas. This changes the pH inside the bag resulting in a color change in the phenol red.

What is phenol used for?

Phenols are widely used in household products and as intermediates for industrial synthesis. For example, phenol itself is used (in low concentrations) as a disinfectant in household cleaners and in mouthwash. Phenol may have been the first surgical antiseptic.
