A commonly used reporter gene is the luciferase gene from the firefly Photinus pyralis. This gene encodes a 61-kDa enzyme that oxidizes D-luciferin in the presence of ATP, oxygen, and Mg(++), yielding a fluorescent product that can be quantified by measuring the released light.

What is the purpose of luciferase?

Luciferase is an enzyme used for bioluminescence by various organisms in nature, most famously the firefly. The scientist produces a construct in which the regulatory region of a target gene is fused with the DNA coding sequence for luciferase (Figure 15.24).

What is the gene code for luciferase?

The luciferase gene was composed of seven exons, coding for 547 amino acids, separated by six introns and spanning 1976 bp of genomic DNA (Fig. 1 and Figure 2). The entire 3086 bp sequence, including primer sequence, was deposited in GenBank, accession # DQ072141.

What is the most common use of luciferase reporter gene?

Luciferase reporter assays are widely used to investigate cellular signaling pathways and as high-throughput screening tools for drug discovery (Brasier et al. 1992, Zhuang et al. 2006).

What is luciferase reaction?

Luciferase is an enzyme. ... The interaction of the luciferase with oxidized (oxygen-added) luciferin creates a byproduct, called oxyluciferin. More importantly, the chemical reaction creates light. Bioluminescent dinoflagellates produce light using a luciferin-luciferase reaction.

Can luciferase be scanned?

The bioluminescence emitted can be detected and amplified using specialized cameras with highly sensitive detection systems [18], revealing the sites and levels of luciferase expression and activity from within a living animal.

Where does luciferase come from?

Luciferase is a light-producing enzyme naturally found in insect fireflies and in luminous marine and terrestrial microorganisms.

Who invented luciferase?

Robert Boyle (1627 - 1691) Important Related Discoveries of the 18^th Century. The Discovery of Luciferin and Luciferase by Raphaël Dubois.

What chemical does the luciferase seek to bind with?

Inside each cell, the newly made enzymes seek out and bind to a chemical called Luciferin. Step 3. The Luciferase Enzyme speeds up a chemical reaction that combines an oxygen molecule with Luciferin to form oxyluciferin.

What is the half life of luciferase?

When placed under the control of estrogen response elements and expressed in human breast cancer T-47D cells, the modified luciferase protein (LUCODC-DA) displayed a functional halflife of 0.84 h compared to 3.68 h for the wild-type enzyme.

How long does luciferase last?

This 36 kDa enzyme is ATP-independent and uses coelenterazine and oxygen as its cofactors. This system provides a so-called long–flash emission, an approximately 30-second burst of 480 nm light. Perhaps the most widely-known luciferase for biomedical purposes is known as Photinus pyralis, or the American firefly.

Is luciferin light sensitive?

Firefly luciferin is sensitive to light, oxygen, and moisture and must be protected.

How do I activate luciferase?

A Luciferase Reporter Assay.

When this protein activates transcription, the cell will produce luciferase enzyme. After the addition of a lysis buffer and a substrate, a luminometer quantifies the luciferase activity. If your protein activates the expression of the target gene, the amount of signal produced increases.

What is luciferase immunoprecipitation system?

Luciferase Immunoprecipitation System (LIPS) assay is a rapid, simple, and sensitive test to detect antibody response to respiratory syncytial virus | FDA. The .gov means it's official.

Why is luciferin called luciferin?

"Luciferin" got its name from the Latin word "lucifer" (meaning "light-bearing"), which is also a source of the word that is sometimes used as a name of the devil.

What is the difference between luciferin and luciferase?

One is a luciferin, or a light-producing substance. The other is a luciferase, or an enzyme that catalyzes the reaction. In some cases, the luciferin is a protein known as a photoprotein, and the light-making process requires a charged ion to activate the reaction.

What is the significance of using luciferase in a bacterial culture?

A particular advantage of luciferases is that most cells are not luminescent so that high signal-to-noise ratios can be achieved. This makes them particularly suited to the analysis of promoters with low activity.

Is luciferase a plasmid?

Luciferase-containing plasmids are commonly used to investigate the effect of regulatory elements, such as promoters, enhancers and untranslated regions, or the effect of mutations of these regulatory elements on gene expression.

Is luciferin harmful to humans?

Luciferin Toxicity

The small size of luciferin also makes it a poor antigen and immune responses to luciferin are unlikely. Luciferin is able to pass the blood brain barrier, the blood placenta barrier and the blood testis barrier, toxicity appears low.

Why is luciferase the best reporter?

Luciferase-based assays are better than other reports because of the following advantages: Quick and real-time measurement. Exceptionally high sensitivity than fluorescent reporters like GFP (10- to 1,000 fold) Range of measurement is wide and dynamic.

How is bioluminescence made?

How it's made. Bioluminescence occurs through a chemical reaction that produces light energy within an organism's body. For a reaction to occur, a species must contain luciferin, a molecule that, when it reacts with oxygen, produces light.

What is a luminometer?

A luminometer is an instrument that measures weak emissions of visible light coming from a sample by means of a photomultiplier tube. ... Light meter is also a broad term but it normally refers to a small device used in photography to measure ambient light.

What animals have luciferin?

• Firefly luciferin is the luciferin found in many Lampyridae species. ...
• Coelenterazine is found in radiolarians, ctenophores, cnidarians, squid, brittle stars, copepods, chaetognaths, fish, and shrimp.

How do humans use luciferin?

Luciferin actually has many applications within the medical field. One of the main ones is bioluminescent imaging, which can be used in vivo (inside a living organism) for whole-body imaging.
